Application

This kit can measure micro free hemoglobin (FHb) content in serum or plasma. 

Detection significance

Plasma free hemoglobin (FHb) refers to the content of hemoglobin in plasma. Usually hemoglobin exists in the red blood cells. When red blood cells are destroyed, hemoglobin will be released. The determination of the content of free hemoglobin is used to reflect the situation of red blood cell damage in hemolytic anemia patients. The increase in free hemoglobin in the plasma generally caused by intravascular hemolysis, globin generation barrier anemia or autoimmune hemolytic anemia.

Detection principle

Ferroheme in hemoglobin molecule have the activity of peroxidase. It can catalyze hydrogen peroxide (H2O2) to release the new ecological oxygen ([O]) which oxidizes phenol and 4-AAP to generate red substance. FHb content can calculated indirectly by measuring the OD value at 510 nm.

Experimental instruments

Test tube, Micropipettor, Vortex mixer, 37℃ water bath/gas bath, Spectrophotometer (510 nm)

Operation steps

Mix fully, incubate in 37℃ water bath for 20 min. Set to zero with distilled water and measure the OD values of each tube at 510 nm with 1 cm diameter cuvette.
